Set clocks ahead for daylight saving time

(AP) ? Spring must be around the corner. It's time to set the clocks forward for daylight saving time.

At 2 a.m. local time Sunday, daylight saving time arrives with the promise of many months ahead with an extra hour of evening light.

You lose an hour of sleep, but make sure to turn the clock ahead ? spring forward ? before heading to bed Saturday night to avoid the panic of a late rise.

It's also a good time to put new batteries in warning devices such as smoke detectors and hazard warning radios.

Some places don't observe daylight saving time. Those include Hawaii, most of Arizona, Puerto Rico, the Virgin Islands, American Samoa, Guam and the Northern Marianas.

Daylight saving time ends Nov. 3.

NKorea warns of more nuclear, rocket activity

SEOUL, South Korea (AP) ? North Korea is warning of future nuclear and rocket efforts that it says are warranted because of new U.N. sanctions over its third nuclear test.

The statement Saturday from an unidentified Foreign Ministry spokesman builds on a torrent of warlike threats from Pyongyang in the wake of the sanctions, which North Korea says are an indication of U.S. hostility.

The sanction, approved Thursday, target North Korea's ruling class.

U.N. diplomats say the sanctions will hurt leader Kim Jong Un. But others argue they may actually bolster him by allowing officials to whip up anti-U.S. sentiment and direct attention away from government failures.

Pyongyang's statement says "the world will clearly see what permanent position" it will take to "reinforce as a nuclear weapons state and satellite launcher" because of the sanctions.

As it'd be poor taste to start singing So Long, Farewell from The Sound of Music, we'll have to settle for a cheery auf wiedersehen and some fond memories of our week in Hannover. Admittedly, this year's show was hardly a vintage for consumer tech news (something of a trend this year, non?), but we certainly saw a few things that made the trip worthwhile. Dan's favorite had to be the eye-tracking AR goggles, while Dana had a soft-spot for those Coby tablets. We're off to load up on chocolate and rindersteak for the flight home -- but you can re-live every nail-biting moment of the CeBIT 2013 if you take a trip past the break.

HTC?s dreadful February shocks even cynics

HTC?s (2498) share price dove 6% yesterday as investors fretted over whether February revenue might drop below NT$12 billion. Well, it did. In fact, it plunged all the way to NT$11.37 billion. This was a -27% decline compared to January? and January revenue was down -28% from December. That January tumble was a pretty normal seasonal drop, but this February decline definitely isn?t. As a sign of that, HTC?s January revenue was down only -7% year-on-year ? meanwhile, February sales were down -44%.

The new HTC One smartphone is about to launch, so it is understandable that carriers might pull back from ordering HTC?s older Android models. But what makes the -44% annualized sales collapse hard to understand is the fact that there are two major new Windows Phones that HTC is now rolling out after light production volumes of December and January.

The high-end Windows Phone 8X and the budget Windows Phone 8S are ramping up widely in Europe and Asia. These two models should be now in the middle of their biggest production volume expansion. How can HTC?s overall volume possibly be down by -44%, particularly since the January sales were not disastrous? This might be a bad omen for the company?s Windows Phone prospects.

There is a lot of buzz about component problems HTC is having. Some analysts have been taking down their early volume estimates for the HTC One in anticipation of small early production batches.

If this sounds familiar, it is. HTC, Nokia (NOK), BlackBerry (BBRY) and some smaller vendors have all started suffering seriously from slow and thin early production runs of major models. This can cause profound damage to the vendor if orders for its older models dry up while operators refuse to commit to major marketing pushes to new models until they get bigger shipments. The danger is that once the real volume production starts, the hot new model is already looking a bit shopworn. Operators have now a smorgasbord of new models to choose from ? Sony?s (SNE) new Xperia is a surprise smash hit in France and the UK in March, usurping the position HTC used to have as a sleek alternative to Samsung (005930).

One thing is clear; HTC One production better ramp up quickly in April even if the March launch is a bit wonky. Huge debuts from Samsung and Apple (AAPL) are already looming in the horizon, ready to suck the oxygen out of the high-end markets in Europe and North America.

Spinal tap -- using cactus spines to isolate DNA

Mar. 5, 2013 ? Isolation of DNA from some organisms is a routine procedure. For example, you can buy a kit at your local pharmacy or grocery store that allows you to swab the inside of your cheek and send the sample for DNA sequencing. However, for other organisms, DNA extraction is much more problematic. Researchers at Desert Botanical Garden in Phoenix, Arizona, have developed a novel procedure that greatly simplifies genomic DNA isolation from cactus tissue.

For members of the family Cactaceae, isolation of genetic material can be difficult due to the presence of polysaccharide-based mucilage content and other secondary compounds. Although important for water storage, these compounds necessitate the use of toxic chemicals and numerous modifications to protocols for DNA extraction. Lead author Shannon D. Fehlberg and colleagues describe a novel method for isolation of DNA using cactus spines in the March issue of Applications in Plant Sciences.

"I had worked with getting DNA out of cactus in the past where you use pieces of the epidermis, but it was messy and difficult to sample. It was also difficult to deal with in the lab because of the mucilage," says Fehlberg. "Now you can snip a spine and, while you have to grind the spine up, it is easy to collect and easy to store and you can follow the manufacturer's protocols for extraction -- simplifying both the field and genetic work."

Considered to be modified leaves, spines contain significantly less mucilage content compared to other tissues commonly used for sampling in Cactaceae. Additionally, removal of cactus spines is less invasive than sampling epidermal tissue, which can damage plants and expose the underlying soft tissue to pathogens.

"Although you can cut a fairly small sample of epidermal tissue, this can be problematic if you are working with living collections or endangered species. Not only is it much easier to clip a spine, it is also more aesthetic and less harmful," comments Fehlberg.

As the cost of DNA sequencing has dramatically decreased, its use has grown exponentially. Because it allows the comparison of individuals within and between populations, DNA sequencing has played an important role in understanding genetic diversity. "For example, in the plant species I'm studying, the species boundaries are not clear," says Fehlberg. "Genetics is important for determining what can be considered a cohesive group. "

Knowledge of genetic variation among populations will provide insight to the persistence of a species and inform conservation efforts. Fehlberg notes, "Genetics is helpful in determining how similar populations are to one another and how connected they are. We're able to use both genetics and biological information to determine which populations are most unique and which are most threatened."

W.Va. House committee votes to weaken EPA standard

CHARLESTON, W.Va. (AP) -- After a contentious public hearing pitting the coal industry against environmental advocates, the West Virginia House Judiciary Committee, in a near unanimous voice vote, advanced legislation that would weaken the state's selenium regulations.

The bill would authorize the state Department of Environmental Protection to conduct a study to determine state-specific guidelines for how much selenium is acceptable in state waters.

If sites are found to have exceeded selenium guidelines, that would no longer be treated as a punishable violation, but would instead trigger additional monitoring.

The U.S. Environmental Protection Agency sets the guidelines for how much selenium is permissible and any change in the state's standards would have to be approved by the agency. If West Virginia changed its standards and the EPA rejected the change, it could lead to litigation.

The EPA has been revising its selenium standards since 2004. An agency spokesman said officials expect to finalize them by the end of this year.

The bill's sponsors contend that the federal regulations are overly restrictive and that selenium has not been proved harmful in West Virginia waterways. They say that West Virginia's fast-moving streams can tolerate higher selenium levels than slow moving rivers and stagnant lakes.

"Our state is better equipped to handle this," said Del. Justin Marcum, D-Mingo, one of the bill's sponsors. "Not a bunch of outsiders coming in telling us how to manage our coal industry."

Del. Clif Moore, D-McDowell, criticized the bill's opponents for impeding economic growth in southern West Virginia and urged the bill's passage.

"I wonder what all these do-gooders were doing a few years ago when McDowell County was literally dying on the economic vine," Moore said.

Selenium is a naturally occurring element that mountaintop removal mining releases into waterways. Studies have found it's harmful to aquatic life, and high-level exposure in humans can damage the kidneys, liver, and central nervous and circulatory systems.

Both supporters and opponents of the bill cited a 2010 study by the state Department of Environmental Protection on selenium's impact on selected fish populations in West Virginia.

Dianne Bady, founder and director of the Ohio Valley Environmental Council, displayed pictures of fish, documented in the study, with massive under-bites and crooked spines and accused the bill's authors of dishonesty and misreading the research.

"House Bill 2579 states the purpose of this bill is to protect state waters," Bady said. "This is a dishonest statement. In reality the purpose of this bill is to allow mountaintop removal companies to pollute West Virginia streams with levels of selenium that are known by the best scientific evidence at this time to cause serious harm to fish and other aquatic life."

Jason Bostic, vice president of the West Virginia Coal Association, said there are four different kinds of selenium and they all have different effects, which are difficult to document.

"For selenium to have an adverse impact it must accumulate in the tissue. Mere exposure to selenium in water is not enough to be able to cause a problem," Bostic said. "Despite searching for an impact, none can be found in West Virginia streams."

Bostic said that the deformities found in the 2010 study are statistically insignificant and have not been verified by subsequent research

The 2010 study found that deformities in fish populations in West Virginia waters were "associated with waterborne selenium exposure."

Randy Huffman, the secretary of the Department of Environmental Protection, acknowledged that the study found fish deformities attributable to selenium, but said that more study was needed.

"We drew some conclusions," Huffman said of the 2010 report. "To caveat that, there is not enough in this study for us to make any conclusive statements."

The bill's supporters pointed to the fact that the EPA selenium regulation placed on streams near coal mines is 10 times stricter than the regulation placed on drinking water. EPA has not commented on how it arrived at its standards, but Tom Clarke, an Environmental Protection department spokesman, said that the distinction is probably due to the differences between aquatic life and humans.

"People are larger and much more complex organisms than some of the organisms living in our streams and therefore are capable of absorbing and ingesting higher concentrations of selenium without being harmed," Clarke said last week. "And another thing is how much water you drink a day is different than a tadpole that is constantly exposed over its whole body."

Hugo Chavez legacy: a wedge between US, Latin America

In December 1994, Miami and the Clinton administration hosted the first Summit of the Americas, an event that drew the leaders of every country from Canada to Chile but Cuba. It was perhaps the zenith of the quest to cast the Western Hemisphere in Washington?s image, with a vast, Arctic-to-Tierra-del-Fuego free-trade area among market economies that banished populism and ostracized Cuba as the lone vestige of a bygone socialism.

Enter Hugo Ch?vez, a red-bereted, self-described Bolivarean revolutionary ? after the George Washington of South America, Sim?n Bol?var ? preaching a very different vision to a struggling but oil-rich Venezuela. Mr. Ch?vez?s model of socialist populism struck a chord that reverberated well beyond Venezuela ? and that sounded the death knell of the thinking, more than a century old, that Latin America had no option but to follow Washington?s lead.

Ch?vez died Tuesday after a long illness that, in recent months, silenced the usually vituperative, blustery, even outrageous leader. He leaves a Latin America much changed from the one he encountered when he first took office as Venezuela's president in 1999.

The change is not so much because Latin America adopted the model Ch?vez espoused, but rather because Ch?vez made it his project to persuade others ? in part by handing out vast amounts of his country's oil revenues ? that alternatives to Washington?s economic and political vision were possible.

?I don?t think it can be overstated how he fundamentally changed relations not just between the US and Venezuela, but how he did the same with US-Latin America relations,? says Eric Farnsworth, vice president of the Council of the Americas and the Americas Society in Washington. ?Ch?vez put meat on the bones of the basic message he spread, which was that there is an alternative for development, [and] you don?t have to follow the example and dictates of the United States.?

News of Ch?vez?s death came the same day the Venezuelan government, increasingly agitated by the president?s worsening condition, accused ?imperialist? enemies ? specifically the United States ? of having infected the president with cancer. The man Chavez designated to succeed him, Vice President Nicolas Maduro, claimed on national television that officials at the US Embassy in Caracas had been involved in conspiracies with Venezuelan military officials to undermine the government and had been expelled.

The US quickly rejected the allegations against US officials, saying such ?fallacious? accusations would make improved relations between the two countries more difficult.

?An assertion that the United States was somehow involved in causing President Ch?vez?s illness is absurd, and we definitively reject it,? said State Department spokesman Patrick Ventrell, in a statement. ?We completely reject the Venezuelan government?s claim that the United States is involved in any type of conspiracy to destabilize the Venezuelan government.?

During his years at the helm of Venezuela, Ch?vez made the pursuit of alternatives to Washington?s vision possible in two key ways, argues Mr. Farnsworth.

First, ?he put his money where his mouth was,? he says, noting that Ch?vez used Venezuela's oil largess to rescue the communist regime of his mentor, Fidel Castro, and to allow other small Latin American countries to discount Washington and its wishes. Ch?vez, Farnsworth says, ?made the fashionable feasible.?

Latin leftists certainly predated Ch?vez, but the economic limitations of their policies eventually clipped their wings: Mr. Castro is the poster boy for that retrenchment from regional dreams. But Ch?vez, sitting atop what are now considered to be the world?s largest oil reserves, used oil revenues in the service of his vision ? even as the Venezuelan economy and the nation's living conditions suffered.

?Previous to Ch?vez, what would a country like Ecuador do, or Nicaragua?? Farnsworth says. Ch?vez?s petroleum diplomacy ?allowed them to chart a different course.?

Second, Farnsworth says, Chavez?s ?outrageousness? ? taking the United Nations stage to label President George W. Bush ?the devil,? or striking up a strategic partnership with Iran ? gave other Latin countries the ?political cover? to do things that were ?not nearly as outrageous? but that nevertheless charted new, non-US-centric directions. Examples include developing strong trade ties with countries other than the US ? Japan or China, for example ? and opposing Washington on the war in Iraq.

Venezuela?s oil sales to China have soared, and Ch?vez signed a $40 billion loan agreement with Beijing that cements China?s access to Venezuelan oil. More worrying still to Washington was how Ch?vez offered Iran a portal into the hemisphere ? and the opening that could provide to a Tehran already embroiled in a covert war with the US.

Ch?vez's ties with other anti-Western leaders who balked at dominance by the world's big powers ? Iran?s Mahmoud Ahmadinejad, Syria?s Bashar al-Assad, even Libya?s Muammar Qaddafi ? certainly rubbed Washington the wrong way. But what set Ch?vez apart was his determination to undermine US influence throughout Latin America and to enlist others to create a united front to that end.

?The rise of Hugo Ch?vez recast the search for regional integration,? says Miguel Tinker Salas, a professor of Latin American studies and a Venezuela specialist at Pomona College in Claremont, Calif. With Ch?vez rocking the boat, the Miami Summit of the Americas' vision of a hemispheric free-trade area faltered, and regional attempts at unity, with no US presence, arose.

?Concentrating the new efforts at economic union and political unity on Latin America diminishes the power of the US in the region, and diminishes the relevance of [Washington-based] institutions like the OAS [Organization of American States],? Professor Tinker Salas says. New collective organizations such as Mercosur, UNASUR, and even ALBA, Ch?vez?s ?Bolivarian Alliance for the Americas,? sprang up. Ch?vez was "a central figure in that,? he says.

But it?s also true that he reached the peak of his influence a few years ago, experts say ? probably when George W. Bush was still president. Since then, Ch?vez?s model of political and economic development for Venezuela has lost much luster.

Moreover, despite his vision of regional unity, Ch?vez was a divisive figure in his own neighborhood, never overcoming testy relations with next-door neighbor Colombia, whose FARC guerrilla fighters he championed.

To illustrate how Ch?vez?s image had tarnished in recent years, some Latin American analysts cite the case of Peru?s president, Ollanta Humala. When Mr. Humala first ran for the job in 2006, he touted his kinship with Ch?vez ? both leftist former army officers ? at every turn. He presented himself as the anti-capitalist who would emulate the populism of Ch?vez and other Latin leftists. He lost that race, but ran again and won in 2011, this time eschewing any admiration for the Ch?vez model.

Since Humala?s election, Peru has enjoyed accelerated economic growth and has moved to join Brazil, Chile, Mexico, and others in combining growth with social improvements without resorting to Ch?vez?s populism or to his antidemocratic concentration of power.

?What the Latin America of the past few years demonstrates is that you don?t have to be a militant populist who is anti-American to accomplish social change and govern with a social conscience,? Farnsworth says.

With Ch?vez?s death, the question for Venezuela and Latin America becomes, ?Will there be Chavismo without Ch?vez?? says Tinker Salas.

For Venezuela, the short-term answer appears to be yes. In elections for state governors in December, Chavista candidates ? several of them leftist former military officers like Ch?vez ? trounced the opposition.

Latin America, on the other hand, has already largely moved on from Ch?vez, though a few leftist populist states still depend on Ch?vez largess. Even so, strains of Ch?vez?s anti-imperialist, region-centric doctrine and his socialist rhetoric are heard in the region's new responses to global and economic challenges, some experts say.

Ch?vez sits with Fidel Castro and ?the sainted Che Guevara? in touching ?a chord in Latin America that is there,? says Charles Shapiro, a former US ambassador to Venezuela who is now director of the Institute of the Americas in San Diego.

But these days, Mr. Shapiro adds, a stronger chord than Ch?vez?s is being struck by Brazil, Chile, Colombia, Peru, Mexico and other countries that are not just talking about poverty, but reducing it.

Facebook has not gotten its fake account problem under control ??yet ? but it appears to be making progress.

In its annual report, Facebook said that the number of duplicate accounts on its network had risen to 5 percent of all accounts in Q4 2012, up from 4.8 percent in Q2 2012.

However, the number of accidentally misclassified accounts and the number of abusive accounts (run by spammers and the like) has declined.

Overall, the total percentage of fake accounts declined from 8.7 percent to 7.2 percent. In whole numbers, the number of duplicate accounts went up from about 45.8 million to 52.8 million; but the total number of fake accounts (which includes abusive and misclassified accounts) declined from 83 million to 76 million over the last six months:

Facebook has therefore deleted or otherwise shut down about 7 million bogus accounts of one sort or another in the last six months, bringing the fake population down from about 83 million in Q2 to 76 million in Q4.

The improvement came after a campaign again fakes, vp/global marketing solutions Carolyn Everson told us back in December. Facebook began its purge of fake accounts and the likes they generated, back in August. After the fake purge began, Lady Gaga lost 65,505 fans, and Facebook's own Facebook page lost 124,919 fake likes, according to data from Likester, the Facebook marketing data company.

The progress is important to Facebook's reputation, even though the actual rate of questionable activity on Facebook is a small percentage of its entire traffic. Advertisers use likes and fan data to target campaigns ? they don't want to waste money advertising on fake accounts.

Facebook has been sued over invalid clicks in the past (the company denies the allegations). There are persistent reports of fake likes coming from the accounts of deceased friends. The system has had flaws in it in the past that have inflated the number of likes on a page. And advertisers on Facebook have grumbled about paying for traffic they believe comes from fakes.

The company is going after bad actors first ? which is why abusive accounts declined. It isn't concerned so much with the majority of duplicate accounts, such as those made for people's pet dogs and cats. So Fido is safe ... for now.

Demolition starts at Florida home where sinkhole devoured man ...

(CNN) ? As a giant red crane plunged into the Florida home where a massive sinkhole swallowed a man whole, pieces of the family?s lives were pushed into public view.

Walls with picture frames on them came crashing down. Baby toys and clothes on hangers were raked across the ground.

A woman wept as an official handed her a framed portrait. Others lovingly salvaged military awards, a pink teddy bear, and an American flag that hung near the house?s front door. The family Bible bore claw marks from the boom crane?s bucket.

Workers started demolishing the blue, one-story home as carefully as they could Sunday to try to salvage belongings for the family of the victim, Jeff Bush.

The delicate process will continue Monday, and crews will clear the debris so engineers can get a better look at the sinkhole and figure out the best way to fill it.

But Bush, 36, probably won?t be recovered. His body remains buried somewhere in the massive sinkhole that stretches 20 feet wide and more than 50 feet deep.

Authorities made the heartbreaking decision to stop the search for Bush after his odds of survival became abundantly clear.

?We just have not been able to locate Mr. Bush, and so for that reason, the rescue effort is being discontinued,? Hillsborough County Administrator Mike Merrill told reporters Saturday. ?At this point, it?s really not possible to recover the body.?

A deafening noise

The family?s nightmare began Thursday night, just as everyone was about to go to sleep.

A deafening noise shattered the peace in house in the Tampa suburb of Seffner.

Jeremy Bush heard his brother scream and ran toward Jeff?s bedroom.

?Everything was gone. My brother?s bed, my brother?s dresser, my brother?s TV. My brother was gone,? he told CNN?s AC360.

Jeremy Bush jumped in the hole and frantically shoveled away rubble. But as the house?s floor further collapsed, a sheriff?s deputy pulled him to safety as his brother remained trapped below.

?I couldn?t get him out,? Jeremy Bush said, weeping. ?I tried so hard. I tried everything I could.?

Jeremy Bush and four others, including a 2-year-old child, were uninjured.

?One step at a time?

After the search for Jeff Bush ended, attention turned to razing the house, which officials warned could collapse at any time.

The demolition crew worked for only a few hours Sunday to give the family time to sift through their belongings, Merrill said.

Once officials get a better view of the sinkhole, ?they can get a sense of what the next step is,? Merrill, the county administrator, said Sunday.

?This is one step at a time, because we really don?t know what we?re dealing with here,? he said.

A major problem in Florida

Sinkholes are a common problem in the state, according to the Florida Department of Environmental Protection.

Florida lies on bedrock made of limestone or other carbonate rock that can be eaten away by acidic groundwater, forming voids that collapse when the rock can no longer support the weight of what?s above it.

Hillsborough County, on Florida?s west coast, is part of an area known as ?sinkhole alley? that accounts for two thirds of the sinkhole-related insurance claims in the state, according to a Florida state Senate Insurance and Banking Committee report.

?So many memories?

The crater that suddenly caved under the Bush house devastated the family that had lived there for generations.

After officials called off the search for his brother?s body, Jeremy Bush told Bay News 9 the family was despondent.

?It?s not just I lost my brother. There are so many memories in this house,? he told the CNN affiliate. ?My wife and her brother and the whole family.

?Every holiday, we gathered at this house. Her grandmother passed away. All the stuff to remember her by is in this house, and we?re losing it all. You can?t replace that. You can?t replace a life being gone.?

Iraq shuts border post after Syrian rebels seize frontier

MOSUL (Reuters) - Iraq shut a border crossing with Syria on Sunday after rebels battling to overthrow Syrian President Bashar al-Assad seized the other side of the frontier post from his army, Iraqi military sources said.

The Iraqi army fired warning shots into the air late on Friday as Syrian insurgents fought government forces in the Syrian town of Yaarabiya and eventually seized the border post.

The fighting so close to Iraq illustrates how Syria's near-two-year conflict could spill over its borders, dragging in neighboring countries and further destabilizing the region.

"Iraqi authorities were ordered to shut off Rabia border crossing until further notice because of the Syrian government's lack of control over the other side of the post," police said.

Syria's insurgents are predominantly Sunni Muslim and are supported by mainly Sunni regional powers such as Turkey and Gulf Arab countries, while Assad belongs to the minority Alawite sect and is backed by Shi'ite Iran.

The conflict has put pressure on the already precarious sectarian and ethnic balance in Iraq. Iraqi Prime Minister Nuri al-Maliki, a Shi'ite Muslim, says his government has a policy of non-interference in Syria, even though his interests are aligned with those of Iran.

Military sources said blast walls now blocked off the border crossing and employees had been evacuated, though both sides of the crossing were calm and there was no sign of Syrian troops or the rebel Free Syrian Army (FSA) forces across the border.

More than 20 Syrian soldiers who fled the fighting into Iraq have been handed over to military intelligence and sent to Baghdad, a military official said.

The Google Play gift card saga continues to roll for eager potential UK based customers, with this latest information appearing on the Google Play support pages. There's no telling at the moment just how long this information has been there, but what it confirms is the amounts we'll -- eventually -- be able to purchase. And it isn't in anyway a surprise. 

It seems that the UK price structure will follow an almost identical numerical scheme to the U.S. cards, with £10, £25 and £50 cards to be available. No £15 one, but that's not a disaster. 

Considering the first signs of a UK launch were seen back in December, the fact we're yet to see these on UK shelves is still somewhat frustrating. Missing the holiday shopping period could be seen as something of a fail on Google's part, but at this point we'll take them when we can get them.

Order in the chaos of a cell membrane

Mar. 1, 2013 ? An explanation has been proposed for the way in which ordered structures arise in cell membranes. Scientists from the Max Planck Institute of Colloids and Interfaces in Potsdam have discovered how complex compounds of sugar and lipids -- known as glycolipids -- order themselves in cell membranes into rafts, namely small, highly organised domains. The arrangement of glycolipids on the surface of plant and animal cell membranes regulates numerous cellular processes. If errors occur in this process, diseases like PNH and BSE can arise.

Lipids, i.e. fats and fat-like substances, arise all over the human body. They are the body's most important energy storage system and are crucial structural components of cell membranes. Compounds formed from complex sugar components and fats are known as glycolipids. Those are vital communicators found in the membranes of every human cell, and constantly exchange information about the type and state of the cell. Numerous metabolic processes depend on glycolipids and their recognition. Even the immune system identifies and combats many pathogens using certain sugar structures located on the surface of the pathogen cells.

Glycosylphosphatidylinositols (GPIs) belong to the group of natural glycolipids. They are found on the surface of plant and animal cell membranes, where they appear either as free molecules or as membrane anchors for various proteins. The arrangement in clusters and their preference for denser and, in part, highly-organised micro-domains in the membrane are seen as essential for the effective functioning of a cell. These minuscule clusters are extremely important for the regulation of many cellular processes, and their malfunction can have very serious consequences. For example, it has been proven that the accumulation, missing or alteration of GPI-anchored molecules can trigger the development of serious diseases like BSE and paroxysmal nocturnal hemoglobinuria (PNH). Scientists at the Max Planck Institute of Colloids and Interfaces in Golm near Potsdam have gained new insight into how GPIs structure themselves in membranes.

Crystalline lipid areas never previously observed in membranes

It was assumed up to now that the arrangement of the GPIs in clusters and rafts was determined by the water-repelling section of the glycolipids embedded in the cell membrane. The chemical structure of the hydrophobic ends is actually responsible for strong interactions with similarly rigid neighbouring molecules. If the number of the molecules that interact with each other is big enough, rigid and partly organised domains may arise like icebergs on the surface of the ocean.

Cristina Stefaniu and her colleagues have now discovered that, in addition to the hydrophobic ends, the large GPI head groups, which contain sugar, mainly contribute to the formation of the rafts. This means that the hydrophylic part of the molecule is able to build strong interactions with the neighbouring GPI molecules. This part of the molecule is located precisely on the boundary between the membrane surface and the liquid medium. "The interactions between neighbouring GPI molecules result in the formation of crystalline orders that have not previously been observed for other membrane lipids," says Cristina Stefaniu.

Hydrogen bonds connect the hydrophylic head groups

The scientists reached this new conclusion about the order in membranes by studying a model molecule. This is a GPI fragment that was synthesised by the groups headed by Peter Seeberger and Daniel Var?n Silva and that imitates the behaviour of entire GPIs. It forms a very thin film, just one molecule thick, on the surface of the water. This so-called monolayer is the simplified model of a half cell membrane which the researchers analysed using synchrotron x-ray scattering. "Surprisingly, the highly ordered structure in the GPI monolayer is predominantly determined by the bulky hydrophilic head groups that connect through hydrogen bonds," says Stefaniu. A hydrogen bond is a relatively weak chemical bond and usually links two molecules through the bonding of a hydrogen atom from one molecule with an oxygen or nitrogen atom from the other molecule. Thus the monolayers of the GPI fragment are characterised by both the order of the hydrophilic lipid chains and the crystalline arrangement of the GPI head groups.

"The molecular lattices observed here have not yet been described for lipid monolayers," says Cristina Stefaniu. "A similar order forms in lipid bilayers if they are stored at temperatures close to zero degrees Celsius." The strong interactions between the head groups can only be disrupted and the molecular lattice destroyed through the addition of a highly concentrated urea solution, which breaks the hydrogen bonds, eliminates the strong interactions of the head groups and destroys the molecular lattice. In addition, the scientists proved that ordered clusters can arise in mixtures of the GPI fragment with typical membrane lipids, which only form unordered films. Thus, the GPIs are able to generate order in the chaos of a membrane. This special skill could be very important for the GPI interactions in real cell membranes.

?So horrific that it?s hard to believe?: Amnesty International launches petition to overturn ?disgraceful? flogging sentence thumbnail

Amnesty International has?launched a petition calling on the Maldivian government to overturn a court ruling sentencing a 15 year-old rape victim to 100 lashes for an unrelated fornication offence.

The story of the girl from Feydhoo in Shaviyani Atoll, who was convicted of premarital sex in the Juvenile Court February 26 and sentenced to 100 lashes and eight months of house arrest, has been reported by media around the world and been widely condemned by international NGOs and embassies.

?It?s so horrific that it?s hard to believe it?s true: a 15 year old rape survivor has been sentenced to 100 lashes for ?fornication? in the Maldives,? stated Amnesty International, which has followed the case since January.

?The traumatised girl was allegedly sexually abused by her step-father for many years. He has since been charged with sexually assaulting a minor. During the investigation however, authorities came across evidence to support separate charges of fornication against the girl for pre-marital sex,? Amnesty stated, demanding the government overturn the ?disgraceful? sentence.

The Maldivian government ? which is currently vying for re-election to the UN Human Rights Council, launched its?campaign in Geneva on February 28 on a platform of ?women and children?s rights and the rights of persons with disability? ? expressed ?deep concern? at the decision to prosecute the girl.

?Though the flogging will be deferred until the girl turns 18, the government believes she is the victim of sexual abuse and should be treated as such by the state and the society and therefore, her rights should be fully protected,? said the President?s Office in a statement.

?The government is of the view that the case merits appeal. The girl is under state care and the government will facilitate and supervise her appeal of the case, via the girl?s lawyer, to ensure that justice is done and her rights are protected,? the statement added.

Amnesty International welcomed the government?s stated commitment, but noted that ?at this stage she is still facing flogging. We need to keep the pressure up to remove flogging as a form of punishment in her case and all cases in the Maldives.?

Foreign Minister Dr Abdul Samad Abdulla meanwhile announced on state broadcaster Television Maldives (TVM) that the girl had post traumatic stress disorder and certain medical issues, according to local media.

?The girl was abused since she was 12 until she turned 15, so multiple times for three years. She was abused by her stepfather. She has said that her baby was murdered. You can imagine the mental status of a child in this situation. She suffers from this condition,? local media reported Dr Samad as saying.

According to Sun Online, Dr Samad said the international community was not fully informed about the reason for the girl?s sentence, which was not due to her abuse by her stepfather, but her confessed fornication with another man.

He also warned that a failure to unite as a country during times of such international criticism would provide opportunities for ?groups that want to harm the country,? Sun reported.

The government?s denouncement of the sentence is meanwhile at odds with its endorsement by the Adaathath Party, members of which make up the majority of the Ministry of Islamic Affairs.

?The purpose of penalties like these in Islamic Sharia is to maintain order in society and to save it from sinful acts. It is not at all an act of violence. We must turn a deaf ear to the international organisations which are calling to abolish these penalties, labeling them degrading and inhumane acts or torture,? the party said in a statement.

?Furthermore, Allah has decreed that expressing disapproval of issues such as this contradicts with faith in Islam,? the statement continued, quoting verses from the Quran.

?Allah has also commanded that we show no kindness when implementing these penalties,? the party added.

Flogging and politics

UN Human Rights Commissioner Navi Pillay in an address to the Maldivian parliament in November 2011 urged the government to issue a moratorium on sentencing women to flogging for extra-marital sex.

?This practice constitutes one of the most inhumane and degrading forms of violence against women, and should have no place in the legal framework of a democratic country,? Pillay stated.

Her comments were condemned by religious groups in the Maldives, with protesters urging authorities to arrest the UN High Commissioner and?gathering outside the United Nations carrying signboards with slogans such as ?Islam is not a toy,? ?Ban UN? and ?Flog Pillay?.

A presidential state apology for allowing Pillay to speak to parliament was one of the five demands of the December 23 coalition [of 2011], a mass gathering in the capital Male? that saw the fractured opposition unite against President Nasheed on the pretext of protecting Islam.

Following the fall of Nasheed?s government on February 7 2012, government-aligned Progressive Party of the Maldives (PPM) MP Abdul Azeez Jamal Abu Bakr withdrew the Pillay apology petition in parliament.

Great White Shark Protections Set To Go Into Effect In California

SACRAMENTO, Calif. ? Great white sharks off the coast of California will likely enjoy greater protection, as state officials consider whether to add the predator to California's list of endangered species.

The California Fish and Game Commission unanimously voted last month to advance the candidacy of the shark for inclusion on the state endangered species list. That means during a one-year study review the shark will receive the same protections as if it were listed as endangered.

The commission was expected to publish last month's decision on Friday, a move that would officially launch the protections, state wildlife officials said.

Great white sharks are internationally protected and already can't be targeted for catch. But the additional restrictions will also ban incidental takes by net fishermen.

"While targeted sport and commercial fishing for white shark has been banned in waters off California since the mid-1990s, there were some exceptions that allowed for incidental take and take associated with research activities," said Marci Yaremko, program manager for state and federal marine fisheries at the state department of fish and game.

Under the enhanced rules, researchers and gillnet fishermen will have to apply to the Department of Fish and Wildlife for permits to tag or accidentally capture a shark or risk criminal prosecution.

Gillnet fishermen have voiced opposition to the shark review, saying the shark population is not in jeopardy and greater protections are not needed.

The shark review is also expected to provide an estimate of the number of great white sharks in state waters.

labeCommenting on ongoing coalition talks between Likud/Beitenu and Bayit HaYehudi, Shas leader Eli Yishai on Thursday morning 18 Adar 5773 explained that he feels Bayit HaYehudi and Yesh Atid are working to keep Shas out of the coalition, and those efforts appear to be succeeding.

Yishai stated that while Shas aspires to be in the coalition, ?we will not betray our hashkafa, our beliefs or our values?.

Yishai writes on his Facebook page that while he does not have first hand information regarding the progress in the talks between the parties, it appears that there is progress and the current situation is one that will not permit Shas to enter the coalition.

Yishai feels the general elections resulted in an untenable situation and ?there are elements who are taking advantage of the situation to advance their agenda at the expense of voters, who are being betrayed.?

He added that ?entering the coalition and receiving ministries, offices and posts are not an end, but a means to serve the tzibur at large and there is no shame in this?, explaining the party?s desire to become a coalition member. He laments the efforts against the Torah world and the inexperience of the new MKs, which he feels is evident in the manner in which they are conducting themselves.

Yishai promises that in or out of the coalition, Shas will continue doing its utmost to serve the tzibur and remain true to its constituents.
